ProQinase’s products & services are designed to support drug discovery for targeting cancer with a focus on protein kinases.

ProQinase sells in-house produced recombinant kinases for enzymatic kinase tests. Currently 239 active protein kinases (including more than 60 mutant kinases and 10 lipid kinases are available as off-the-shelf products. In addition, additional protein kinase variants (inactive or non-activated) are available from ProQinase. Kinases can also be produced as custom-tailored proteins according to your requirements. Moreover, ProQinase offers recombinant substrates for biochemical kinase activity assays.

 Biochemical Kinase Assay Services

ProQinase offers different types of compound testing in biochemical kinase tests with protein kinases and lipid kinases. Our highly flexible free choice kinase assay projects include high throughput screening (HTS) of large compound libraries against one target, and selectivity profiling against several or all kinases which are available at ProQinase (358 protein kinases and 10 lipid kinases). Kinase tests can be performed either as single concentration measurements or as IC50 determinations.
In addition to free choice kinase assay projects the Kinase WholePanelProfiler (358 protein kinases) and the Kinase WholePanelProfilerPlus (358 protein kinases and 10 lipid kinases) allow profiling test compounds against broad panels of kinases.
Moreover, ProQinase offers to test your compound(s) in predefined Kinase MutantProfiler comprising different mutants and the wildtyp form of a given kinase and including a side-by-side comparison with reference inhibitors.
The KinaseFinder is offered as a standardized service package which enables to identify protein kinases that phosphorylate a protein or peptide of interest.
The Kinase SubstrateFinder is another standardized service package that allows identification of protein kinase substrates for biochemical protein kinase activity assays.

ProQinase offers several types of cell-based assay services to characterize compounds on the cellular level for targeting cancer. On the one hand, we offer cellular phosphorylation assays   to assess the inhibitory effect of compounds on a specific kinase (currently established for 22 different kinases). On the other hand, we offer various sophisticated phenotypic cell-based assays, such as (i) the spheroid-based angiogenesis assay to evaluate the angiogenic or anti-angiogenic potential of compounds, (ii) the cell migration assay which allows to identify the impact of compounds on the motility of various cell lines, (iii) the cell proliferation assay to investigate the effect on tumor cell growth and (iv) the soft agar assay (or clonogenic assay) which characterizes compounds for their ability to inhibit cell transformation.
In all types of cell-based assays testing can be performed in screening mode (using only one or a few concentrations of the test compounds) or as IC50 determination.

ProQinase has established a large panel of subcuteanous xenografts and orthotopic tumor models for in vivo testing. All orthotopic tumor studies are performed with luciferase transfected tumor cell lines allowing bioluminescene imaging.  Moreover, metastasis tumor models are available for in vivo testing of lead compounds. ProQinase's unique spheroid-based in vivo angiogenesis assay allows in vivo testing of the anti-angiogenic effect of compounds or antibodies against functional human blood vessels in mice.
